Condenser Aperture Setting

NOTE: An aperture is selected at each position of the aperture holder. There is no "out" position for the condenser aperture. However, the objective aperture has an out position. The normal condenser aperture position is 150 µm, or the second largest spot.

NOTE: For normal imaging, a SPOT SIZE of 1 and the second condenser aperture (150 µm) are optimal.

1. Set to Page 1 by engaging the PAGE button in the right drawer. VIEW
2. Set MAG = 5000X using the SELECTOR knob. VIEW
3. Focus illumination to a small spot using the BRIGHTNESS knob. VIEW

4. Center illumination using the SHIFT X,Y on the main panel and the black dot on the fluorescent screen as a reference.

5. Spread the illumination with the BRIGHTNESS knob. VIEW

6. If the illumination is not concentric to the center of the screen, move the illumination with condenser aperture drives in the direction of deflection.

   

7. Focus the illumination to a spot using the BRIGHTNESS knob.

   

8. Re-center illumination with the SHIFT X, Y on the main panel.

9. Repeat steps 3 - 8 until the illumination converges to the center when focusing/defocusing with the BRIGHTNESS knob.
